(Medical-NewsWire.com, October 28, 2012 ) San Francisco, CA- When it comes to patients who experience chronic diabetic peripheral neuropathic pain, also known as DPNP, there is little difference between amitriptyline duloxetine and pregabalin. However, a recent study shows that the latter does improve sleep, while the former is useful for daytime functioning.
The contrasting capability was noted by research published online in Diabetes Care.
Julie Boyle, PhD at the University of Surrey in the U.K. and her colleagues conducted a double-blind and randomized study. The study utilized 83 type 1 and type 2 diabetes patients who reported suffering from DPNP.
The study ran for 14 days of low-dose therapy, then higher-dose days of the same length. The three groups were amitriptyline, duloxetine, or pregabalin. The impact of polysomnographic sleep, daytime function, and quality of life.
Researchers concluded that all three medications were capable of pain relief compared to placebo, with no significant difference being observed. Pregabalin was noted to improved sleep in its group. Duloxetine increased waking function. It also lowered to total sleep time of its users. It also improved nervous system arousal and sensory motor skills, despite lowering overall sleep time. Pregabalin had more adverse effects in a number of those studied compared to other drugs.
"In conclusion, amitriptyline, duloxetine, and pregabalin were equally effective analgesic medications in patients with DPNP. Daytime function was relatively unaffected by drug treatment, and all three drugs were well tolerated," the authors write. "In this short, 28-day dosing study, there was no evidence of improved quality of life (Short Form 36) even with the sleep enhancement observed with pregabalin."
The study was funded by an investigator-led research grant awarded by Pfizer; two authors disclosed potential financial conflicts of interest.
